Болезнь Альцгеймера (БА) — это наиболее распространенное нейродегенеративное заболевание, которое вызывает потеряю памяти преимущественно у лиц пожилого и старческого возраста [<xref ref-type="bibr" rid="cit1">1</xref>]. БА является одной из наиболее распространенных причин смерти, наступающей в течение 5–12 лет после появления первых симптомов. Современные лекарственные препараты, одобренные организацией FDA (Food and Drug Administration) для лечения БА (мемантин, такрин, ривастигмин, галантамин, донепезил), не позволяют существенно замедлить прогрессирование заболевания [<xref ref-type="bibr" rid="cit2">2</xref>] и обладают широким спектром побочных эффектов [<xref ref-type="bibr" rid="cit3">3</xref>]. Ввиду отсутствия эффективного лечения, прогрессирующей заболеваемости в мире, а также значительных трат на поддержание жизнедеятельности пациентов, поиск эффективных и безопасных потенциальных терапевтических соединений для лечения БА приобретает все большую значимость на фоне глобальной тенденции к увеличению продолжительности жизни населения. Перспективными нейропротекторами, обладающими низкой иммуногенностью и высокой физиологической активностью, являются пептидные биорегуляторы. В частности, для терапии БА представляется целесообразным изучение активности коротких пептидов EDR и KED, синтезированных на основе анализа аминокислотного состава экстрактов, выделенных из тканей головного мозга и сосудов крупного рогатого скота соответственно.</p></sec><sec><title>ЦЕЛЬ ИССЛЕДОВАНИЯ</title><p>ЦЕЛЬ ИССЛЕДОВАНИЯ. Оценить эффективность коротких пептидов в in vitro и in vivo моделях болезни Альцгеймера с целью разработки средства для лечения и профилактики данной патологии.</p></sec><sec><title>МАТЕРИАЛЫ И МЕТОДЫ</title><p>МАТЕРИАЛЫ И МЕТОДЫ. В качестве in vitro модели БА использовали модель амилоидной синаптотоксичности — культура нейронов, выделенных из мозга новорожденных мышей дикого типа, с добавлением синтетических олигопептидов Aβ. Экспериментальные группы были сформированы следующим образом: 1-я — контроль (без Aβ); 2-я — контроль с добавлением Aβ; 3-я — добавление Aβ и пептида EDR (200 нг/мл); 4-я — добавление Aβ и пептида KED (200 нг/мл). Далее с помощью конфокального микроскопа ThorLabs и программного обеспечения «ThorLabs» (USA) при 100-кратном увеличении (Olympus UPlanSApo) с разрешением 1024 × 1024 получали серии микрофотографий нейронов, которые использовали для анализа морфологии дендритных шипиков с помощью программного обеспечения NeuronStudio software package согласно [<xref ref-type="bibr" rid="cit4">4</xref>]. В качестве in vivo модели БА использовали трансгенную линию мышей 5xFAD-M, которую выводили путем кроссбридинга мышей линии 5xFAD и Thy1-GFP. Экспериментальные группы были сформированы следующим образом: 1-я — контроль (линия мышей без патологии, которым вводили физиологический раствор); 2-я — мыши линии 5xFAD-M (модель БА), которым вводили физиологический раствор; 3-я — мыши линии 5xFAD-M, которым вводили пептид EDR; 4-я — мыши линии 5xFAD-M, которым вводили пептид KED. Короткие пептиды вводили животным внутрибрюшинно в возрасте с 3 до 5 месяцев ежедневно. В возрасте 5 месяцев животным проводили транскардиальную перфузию, извлекали головной мозг и готовили фиксированные срезы мозга толщиной 40 мкм на вибратоме. С помощью конфокального микроскопа ThorLabs и программного обеспечения «ThorLabs» (USA) получали серии микрофотографий нейронов и проводили морфометрический анализ дендритных шипиков.</p></sec><sec><title>РЕЗУЛЬТАТЫ</title><p>РЕЗУЛЬТАТЫ. При оценке эффективности коротких пептидов в in vitro модели БА было показано, что пептид EDR в концентрации 200 нг/мл повышал количество наиболее функциональных грибовидных шипиков нейронов (на 71%) в первичной культуре нейронов мышей в условиях амилоидной синаптотоксичности до уровня нормы. Пептид KED в концентрации 200 нг/мл повышал количество грибовидных шипиков в модели БА на 20%. При оценке эффективности коротких пептидов в in vivo модели БА было показано, что ежедневное внутрибрюшинное введение пептидов EDR и KED мышам линии 5xFAD-М в возрасте с 3 до 5 месяцев в концентрации 400 мкг/кг способствовало статистически значимому увеличению количества грибовидных шипиков в СА1 области гиппокампа соответственно на 25% и 27%. Под действием пептидов увеличивалась и общая плотность дендритных шипиков на 13% и 22% соответственно.</p></sec><sec><title>ВЫВОДЫ</title><p>ВЫВОДЫ. Полученные результаты свидетельствуют о том, что короткие пептиды EDR и KED обладают нейропротекторным эффектом в модели БА, который заключается в улучшении функционального состояния нейронов. Необходимо провести дальнейшие исследования для выявления механизма нейропротекторного действия коротких пептидов EDR и KED при БА, которые позволят приступить к разработке лекарственного средства для профилактики и лечения данной патологии.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>BACKGROUND</title><p>BACKGROUND. Alzheimer’s disease (AD) is the most common neurodegenerative disease that causes memory loss predominantly in eldely and senile people [<xref ref-type="bibr" rid="cit1">1</xref>]. AD is one of the most common causes of death within 5–12 years after the first detected symptoms. Modern drugs approved by the FDA (U.S. Food and Drug Administration) for the AD treatment (memantine, tacrine, rivastigmine, galantamine, donepezil) do not significantly slow down the disease progression [<xref ref-type="bibr" rid="cit2">2</xref>] and have a wide range of side effects [<xref ref-type="bibr" rid="cit3">3</xref>]. In view of the lack of effective treatment, the rising global prevalence of AD and significant costs of patient life support, finding effective and safe potential anti-AD drugs is becoming increasingly critical in the context of a global trend towards longer life expectancy. Peptide bioregulators are promising neuroprotectors with low immunogenicity and high physiological activity. In particular, AD therapy may benefit from studying the activity of EDR and KED short peptides obtained by analysis of amino acid composition of extracts isolated from cattle brain and vessels, respectively. AIM. To evaluate the efficacy of short peptides in in vitro and in vivo models of Alzheimer’s disease with a view to developing a drug for the treatment and prevention of this pathology.</p></sec><sec><title>MATERIALS AND METHODS</title><p>MATERIALS AND METHODS. A neuronal culture isolated from the brain of newborn wild-type mice, with the addition of synthetic oligopeptides Aβ (the amyloid synaptotoxicity model) was used as the in vitro model of AD. The study groups were as follows: Group 1: control (without Aβ); Group 2: control with the addition of Aβ; Group 3: the addition of Aβ and EDR peptide (200 ng/mL); Group 4: the addition of Aβ and KED peptide (200 ng/mL). Series of neuron microphotographs were obtained using the ThorLabs confocal microscope and ThorLabs software (USA) at the 100-fold magnification (Olympus UPlanSApo) with resolution 1024×1024, which were used for dendritic spine morphology analysis using NeuronStudio software package according to [<xref ref-type="bibr" rid="cit4">4</xref>]. The transgenic line of 5xFAD-M mice derived by cross-breeding of 5xFAD and Thy1-GFP mice was used as an in vivo model of AD. The experimental groups were as follows: Group 1: control (a line of healthy mice, which received the injections of physiological solution); Group 2: 5xFAD-M line of mice (AD model), which received the injections of physiological solution; Group 3: 5xFAD-M line of mice (AD model), received the injections of EDR peptide; Group 4: 5xFADM line of mice (AD model), received the injections of KED peptide. Short peptides were intraperitoneally injected in mice aged from 3 to 5 months on a daily basis. At the age of 5 months, the animals underwent transcardial perfusion followed by brain extraction; fixed brain slices (40 μm thick) were prepared using a vibratome. Series of neuron microphotographs were obtained using the ThorLabs confocal microscope and ThorLabs software (USA), followed by morphometric analysis of dendritic spines. RESULTS. EDR peptide at the concentration of 200 ng/mL increased the number of the most functional mushroom spines of neurons (by 71%) to normal levels in the primary neuronal culture under amyloid synaptotoxicity conditions (in vitro AD model). KED peptide at the concentration of 200 ng/mL increased the number of mushroom spines by 20% in the AD model. Daily intraperitoneal injection of EDR and KED peptides at a concentration of 400 μg/kg to 5xFAD mice (in vivo AD model) aged from 3 to 5 months contributed to statistically significant increases in mushroom spines in the CA1 area of the hippocampus by 25% and 27%, respectively. These peptides contributed to the increase in the total dendritic density by 13% and 22%, respectively.</p></sec><sec><title>CONCLUSIONS</title><p> CONCLUSIONS. The obtained results demonstrate that EDR and KED short peptides have a neuroprotective effect in the AD model, manifested by an improved functional state of neurons.
